تُعَدّ الحلبة -graecum من النباتات البقولية ذات الأهمية الطبية والغذائية، مما يستدعي تطوير طرق فعالة لإكثارها مخبرياً باستخدام تقنية زراعة الأنسجة النباتية. هدفت هذه الدراسة إلى تقييم تأثير منظِّمات النمو النباتية على الإكثار الخضري واستحداث الكالس من الحلبة صنف كناكر. تمت زراعة البذور بعد تعقيمها على وسط MS، ثم نُقلت النموات الناتجة إلى أوساط مزودة بتراكيز مختلفة من ‎ 2.0 ،1.5 ،1.0 ،0 :BAP ‎مغ/ل⁻¹، ومن ‎ 0.5 ،0.25 ،0.1 ،0 :NAA مغ/ل⁻¹.

أظهرت النتائج أن تركيز 2 مغ/ل⁻¹ من BAP حقق أعلى معدل في متوسط طول النبات، وعدد الأفرع، وعدد الأوراق: 6.34 سم طولاً، 7.51 فرع/نبات، 14.63 ورقة/نبات على التوالي، كما تفوق تركيز 0.5 مغ/ل⁻¹ من NAA في تعزيز النمو، حيث سجل أعلى معدل لطول النبات وعدد الأفرع والأوراق، وكان التفاعل بين التركيزين 0.5 مغ/ل⁻¹ NAA و2.0 مغ/ل⁻¹ من BAP هو الأفضل في تحقيق أعلى متوسط لجميع المؤشرات المدروسة.

في مرحلة استحداث الكالس، بينت النتائج أن إضافة 1 مغ/ل⁻¹ من D -2,4 إلى وسط MS أدى إلى استحداث الكالس بنسبة 100 %، بينما لم يُلاحظ أي تكوين للكالس في غياب منظمات النمو. تم تسجيل أعلى وزن طري وجاف للكالس عند التركيز ذاته عند عمر 15 يوماً، مما يشير إلى الدور الحاسم للأوكسينات في تحفيز الاستحداث.

تؤكد هذه الدراسة أهمية التفاعل بين BAP و NAA في تحسين الإكثار الخضري للحلبة، مما يوفر بروتوكولاً عملياً يمكن تطبيقه في الزراعة المخبرية لتعزيز إنتاج هذا النبات ذي القيمة الطبية العالية.

Fenugreek -graecum is a significant leguminous plant with medicinal and nutritional value, necessitating the development of efficient propagation methods through plant tissue culture techniques. This study aimed to evaluate the effects of plant growth regulators on the micropropagation and callus induction of the “Kanaker” variety of fenugreek. Sterilized seeds were germinated on Murashige and Skoog (MS) medium, and the resulting shoots were transferred to media supplemented with varying concentrations of BAP: 0, 1.0, 1.5, and 2.0 mg•L-1 and NAA: 0, 0.1, 0.25, and 0.5 mg•L-1.

The results indicated that 2.0 mg•L⁻¹ BAP yielded the highest averages in plant length, number of branches, and number of leaves: 6.34 cm length, 7.51 branches/plant-1 and 14.63 leaves/plant respectively. Additionally, 0.5 mg•L⁻¹ NAA excelled in promoting growth, recording the highest values for stem length, branch number, and leaves number. Moreover, the interaction between 0.5  mg•L⁻¹ NAA and 2.0 mg•L⁻¹ BAP proved to be the most effective treatment in achieving the highest averages across all studied parameters.

For callus induction, the results indicated that supplementing MS medium with 1 mg•L⁻¹ of 2,4-D resulted in 100% callus formation, whereas no callus formation was observed in the absence of plant growth regulators. The highest fresh and dry weights of callus were recorded at the same concentration at 15 days, highlighting the crucial role of auxins in stimulating induction.

This study underscores the significance of the interaction between BAP and NAA in enhancing fenugreek micropropagation, providing a practical protocol for cultivation to improve the production of this valuable medicinal plant.
